DOWNSTREAM BIOPROCESSING

Remove Endotoxin. Preserve the Product.

Endotoxin removal is a persistent challenge in downstream bioprocessing, particularly for products manufactured using Gram-negative bacterial expression systems.

The challenge is not simply removing LPS. It is doing so without losing the valuable biological product being purified.

PCL nanoparticles bind endotoxin while demonstrating comparatively low affinity for proteins under the conditions investigated.

In peer-reviewed research, PCL nanoparticles achieved >99% LPS removal with >99% protein recovery across a reported protein concentration range of 20–1,000 µg/mL.

When compared with five commercially available endotoxin-removal products under the study conditions, the nanoparticle and membrane systems demonstrated approximately 1.25- to 30-fold greater efficiency.

Market Opportunities

Recombinant Protein Manufacturing

Recombinant protein production often requires dedicated downstream steps to reduce residual endotoxin before final formulation. Selective endotoxin capture while maintaining protein recovery could improve purification efficiency. A nanoparticle-embedded membrane could provide an alternative or complementary polishing approach for suitable recombinant protein manufacturing processes.

Biologic & Biosimilar Manufacturing

Biologic and biosimilar manufacturing places increasing emphasis on downstream efficiency, product recovery, and consistent impurity control. Selective endotoxin removal could add a targeted polishing capability for appropriate process streams where maintaining valuable product yield while reducing residual endotoxin is an important manufacturing consideration.

Vaccines & Antigens

Recombinant antigens and other biologically produced vaccine components may require stringent purification and impurity-control strategies before formulation. Membrane-based endotoxin capture could provide an additional selective polishing step for suitable vaccine manufacturing processes where endotoxin reduction and preservation of the target biological product are both important.

Enzymes & Fermentation-Derived Products

Industrial and pharmaceutical enzymes, proteins, and other fermentation-derived biomolecules can encounter endotoxin contamination during production and downstream processing. Selective membrane-based endotoxin removal could simplify final purification for appropriate products while helping preserve recovery of valuable biomolecules produced through bacterial or other fermentation-based manufacturing systems.

Research Reagents & Life-Science Products

Purified proteins, enzymes, antibodies, and other life-science reagents can be highly sensitive to residual endotoxin contamination. A selective endotoxin-polishing technology could strengthen product quality while supporting manufacturing recovery for research-product suppliers producing reagents used in sensitive analytical, cellular, and experimental applications.

Contract Manufacturing & Bioprocess Platforms

Contract manufacturers and bioprocess technology providers continually evaluate modular, single-use technologies that can integrate into existing downstream workflows. A nanoparticle-embedded membrane could be developed as an endotoxin-polishing cartridge or membrane module, providing a flexible additional purification step for compatible biopharmaceutical manufacturing processes.
